课题组拟利用自主开发的新生RNA全长测序技术和单细胞核全长转录组测序技术,结合多种遗传生化和分子生物学手段,从RNA转录、剪接、终止、加A和降解等多个层次研究RNA转录加工调控水稻逆境适应和氮磷营养利用效率的分子机制。

主要学术成果:
长期致力于植物RNA加工及其调控生长发育和环境响应机制的研究,以第一作者在Nature Plants(排名第一,两篇)、Plant Cell(排名第一)、Nature Protocols(排名第二)和Genome Biology(排名第三)上发表多篇研究论文。主要学术成果包括:
(1)通过开发和利用植物新生RNA全长测序技术FLEP-seq,深入研究了植物共转录和转录后RNA剪接加工机制,发现植物普遍存在转录后剪接现象,转录后剪接抑制介导的染色质滞留在植物基因表达调控中发挥重要作用;
(2)利用FLEP-seq深入研究了植物polyA长度调控机制,发现花粉和种子中polyA长度受到特异性调控;
(3)揭示大豆DCL2依赖的22-nt小RNA加工及其调控种皮颜色的机制;
(4)合作开发了植物单细胞核新生RNA全长测序技术,实现在单细胞水平同时检测基因表达和内含子剪接变化动态。
